A DoS is caused by usually a 'botnet' of infected computers around the world sending thousands to maybe millions of packets per second to a server for example. These packets are usually TCP SYN, which when sent to the recieving server cause it to open a session. Each session uses resources (such as CPU/RAM and bandwidth). If you send enough TCP SYNs to the server it will crash, and depending on the size of the network link it may become over utilized and become unusable for anyone else on the network.
